The Turtles respond immediately when April calls for help after a city block is overrun by Kraang robots, and the first mission begins with a direct goal to fight through the streets and stop the invaders from advancing. Each area opens with clear enemy placements, hazards, and checkpoints that must be cleared. Short messages from April or Splinter indicate which path to take or which enemies to prioritize. Every stage starts directly with a simple objective such as defeating all foes, protecting civilians, or reaching the next key location.

Combat relies on each Turtle’s unique weapons and abilities. Leonardo swings his swords, Michelangelo uses nunchucks, Donatello attacks with his bo staff, and Raphael strikes with sai. Moves can be chained into combos, and special attacks consume energy to deal area damage or hit multiple enemies at once. Some sections require dodging projectiles, jumping across broken rooftops, or breaking obstacles to reach the next area. Every hit, block, or dodge shows immediate results, making it clear when enemies are weakened or defeated.

Later missions introduce tougher enemies, larger groups, and mini-bosses with stronger attacks or shields that require specific strategies. New areas unlock, including sewer tunnels, rooftops, and warehouses with environmental hazards like collapsing beams or conveyor belts. Upgrades for weapons and Turtle stats become available as missions are completed, helping handle stronger opponents. Short dialogue sequences indicate the next target or alert the Turtles to hidden threats. The campaign ends after defeating the main villain, closing once the city block is cleared and the Turtles restore safety.
